Inside This Application Note
Explore Ricoh Biosciences' standardized workflow for receiving, expanding, and characterizing custom iPSC lines, including karyotyping, pluripotency marker analysis, and trilineage differentiation testing using the CIRM CW50065 reference line.
- Immunofluorescence staining of the CIRM CW50065 line confirmed 94.5% OCT4-positive and 82.2% NANOG-positive cells, with flow cytometry further showing 92.2% OCT4, 98.9% SSEA-4, and 95.8% TRA-1-60 positivity.
- Directed trilineage differentiation of the CW50065 line for 6 days showed upregulation of three ectodermal markers, one mesodermal marker, and two endodermal markers across three iPSC seeding densities.
- G-banding karyotype analysis of the CW50065 line confirmed a stable 46,XX apparently normal human female karyotype at both passage 16 and passage 30.
