Review a detailed, day-by-day workflow for culturing iPSC-derived excitatory neurons with human primary astrocytes in 96-well spheroid format for calcium flux transient measurement using FDSS or FLIPR instrumentation.
- Neurons are co-cultured with human primary astrocytes at a 1:1 ratio of 8,000 cells each per well, plated in 96-well ultra-low attachment spheroid microplates compatible with FDSS and FLIPR readout systems.
- The protocol spans from Day -7 through Day 28+, transitioning from Medium N(G2P) to Neurobasal Plus maturation medium at Day 7, with twice-weekly half-medium changes for 5 to 6 additional weeks.
- Each Quick-Neuron Excitatory kit provides more than 1.0 million viable cells, sufficient to plate 60 internal wells of one 96-well plate, with edge wells filled with PBS to minimize evaporation artifacts.
