Modeling Alzheimer's Disease with ELISA and Off-the-Shelf Patient iPSC-Derived Neurons

Measure Alzheimer's disease biomarkers in off-the-shelf patient iPSC-derived excitatory neurons, with ELISA-confirmed elevation of Aβ42, Aβ40, total Tau, and p-Tau relative to age-matched healthy controls.

Inside This Application Note

Review ELISA biomarker data from APOE4 patient and healthy control iPSC-derived excitatory neurons cultured for up to 6 weeks, including AD phenotype confirmation and β-secretase inhibitor dose-response results.

  • All four AD biomarkers — Aβ42, Aβ40, total Tau, and p-Tau — showed higher values in the AD patient line relative to the healthy control line at the end of week 6, demonstrating that iPSC-derived neurons display known phenotypes of AD
  • Increasing doses of β-secretase inhibitors LY2886721 and BSI-IV resulted in a corresponding decrease of Aβ42 and Aβ40 in AD patient neuron culture supernatants after 6 weeks of culture
  • Neurons sourced from CIRM-derived APOE4 carrier Alzheimer's disease patient iPSCs and differentiated using Quick Tissue™ technology, available off-the-shelf without requiring in-house iPSC reprogramming or differentiation
